Age-Related Changes in the Enteric Nervous System

چکیده

As we enter the 21 century, the segment of the population that is the most rapidly expanding is that comprised of individuals 85 years of age and older. Dysfunctions of the gastrointestinal (GI) system, including dysphagia, constipation, diarrhea, and irritable bowel syndrome are more common complaints of the elderly, yet our knowledge of the aging GI tract is incomplete. Compared to the rapid advances in the neurobiology of aging in the CNS, the understanding of age-related changes in the enteric nervous system (ENS) is poor. In this brief review I recap experiments that reveal neurodegenerative changes, and their functional correlates, in the ENS of mice, rats and guinea pigs. Clinical literature seems indicative of similar structural and functional age-related changes in the human ENS. Current studies that address the mechanisms underlying agerelated changes in the ENS are introduced. The future directions for this field include physiological and pharmacological studies, especially at the cellular and molecular level. Research in the aging ENS is poised to make major advances, and this new knowledge will be useful for clinicians seeking to better understand and treat GI dysfunction in the elderly.

Loss of UCHL1 promotes age-related degenerative changes in the enteric nervous system

UCHL1 (ubiquitin carboxyterminal hydrolase 1) is a deubiquitinating enzyme that is particularly abundant in neurons. From studies of a spontaneous mutation arising in a mouse line it is clear that loss of function of UCHL1 generates profound degenerative changes in the central nervous system, and it is likely that a proteolytic deficit contributes to the pathology. Here these effects were found...

The Influence of Inflammation and Nerve Damage on the Neurochemical Characterization of Calcitonin Gene-Related Peptide—Like Immunoreactive (CGRP-LI) Neurons in the Enteric Nervous System of the Porcine Descending Colon

The enteric nervous system (ENS), localized in the wall of the gastrointestinal tract, regulates the functions of the intestine using a wide range of neuronally-active substances. One of them is the calcitonin gene-related peptide (CGRP), whose participation in pathological states in the large intestine remains unclear. Therefore, the aim of this study was to investigate the influence of inflam...

Aging and neural control of the GI tract: III. Senescent enteric nervous system: lessons from extraintestinal sites and nonmammalian species.

Functional changes in GI motility associated with advanced age include slowing of gastric emptying, decreased peristalsis, and slowing of colonic transit. These changes appear to be associated with region-specific loss of neurons and impaired function. The mechanism(s) underlying physiological aging are likely to be multifactorial. Alterations in specific signal transduction pathways have been ...

SARS‐CoV2 Neuroinvasion Potential; Narrative Review

The outbreak of severe acute respiratory syndrome (SARS ‐ CoV-2) in December 2019 has led to the epidemic of Covid-19. Recent clinical studies have shown that parts of the nervous system; Cerebrospinal fluid (CSF) are involved in SARS-CoV-2 infection, which is associated with a variety of neurological manifestations such as headache, dizziness, and olfactory and gustatory disorders. This study ...
